It mimics a natural brain growth factor to nudge the hippocampus into making new neurons and to raise BDNF, the signal tied to learning and memory, without the side effects of the full-size molecule.

How it works

P21 is a four-amino-acid mimetic of ciliary neurotrophic factor (CNTF). In animal work it raises BDNF, the brain's main growth-and-repair signal, through the TrkB and CREB pathway, while blocking the LIF and STAT3 signaling that the larger CNTF molecule overactivates.

Where it acts

Its headline effect is in the dentate gyrus of the hippocampus, the region that makes new neurons in adults. P21 is studied for boosting that neurogenesis, maturing the new cells, and restoring synaptic proteins, which is why it is grouped with learning and memory.

What people notice

In rodent studies it improved short-term and spatial memory and lowered anxiety-like behavior. Human reports are anecdotal, describing clearer thinking and better recall that builds over weeks. It is not a same-day stimulant.

How to take it.

  1. Reconstitute into a spray bottle

    Add bacteriostatic water to the vial (a 10mg vial + 2mL = 5mg/mL), swirl gently, don't shake, then draw it into a small intranasal spray bottle so each pump delivers a known dose.

  2. Prime the pump

    With a fresh bottle, spray a few times into a tissue until a fine, even mist comes out. This makes each later spray a consistent amount, usually around 100–250 mcg per spray depending on your mix.

  3. Spray per nostril

    Tilt the head slightly forward, insert the tip, and give one measured spray into each nostril while breathing in gently. Don't sniff hard; you want it to coat the nasal lining, not run down the throat.

  4. Alternate nostrils & store cold

    Rotate which nostril you start with across doses, wipe the tip, and keep the bottle in the fridge between uses. Use a reconstituted bottle within a few weeks.

Typical P21 dose: research reports commonly run roughly 0.5 mg to 1 mg once daily, often in the morning, in blocks of a few weeks rather than continuously. Intranasal doses sit at the lower end because nose-to-brain delivery is efficient.
